I loved The New Tetris for N64. It had a "block" mechanic that gave bonus points if 4 blocks were formed into a 4x4 grid. Has this mechanism been used in any more recent Tetris games?

Sorry that this does not answer your direct question, but I enjoyed site TetrisFriends, which has many different modes, quests and challenges built around the Tetris game. May be you'll like the variety too (though you'll need a browser to try it, so it won't work on N64).

In Tetris Worlds there's a mode called Square Tetris

Give Tetris Attack (SNES) a shot, or any of its logical equivalents, like Planet Puzzle League (DS) and Pokemon Puzzle League (N64).
You can implement combos of complication several magnitudes higher than just 4x4 blocks, including chains that gather weight over time, and sequences that require perfect timing to execute.
